Define S or R6?

I'm not sure which one to go with, both the Define S and R6 have good airflow, simple look and watercooling support. What are the differences between these cases that would make someone buy one over the other?

Depends on your plan. Go to FD's website and download the manual for each case. They have about 15 pages at the end of the manual that showcase all fan and radiator options including limitations to each. Very informative.
